They result from the antibacterial activity and pharmacokinetic characteristics of mezlocillin. They take into account both the clinical studies that the drug has given and its place in the range of antibacterial products currently available.
- They are limited to severe infections caused by the organisms defined above as sensitive and resistant to aminopenicillins (ampicillin and related), in particular in their manifestations:
. septicemic and endocardial
. meningeal (good passage of the meningeal barrier),
. renal and urogenital
. gynecological and obstetrical,
- Official recommendations regarding the proper use of antibacterials should be taken into account.

- In the subject with normal renal functions:
. Adults : The average dose is 240 mg / kg / day, which can be increased to 300 mg / kg / day. The daily dose is administered at least in 3 IV injections spaced 8 hours apart.
. Children and Infants : The average dose is 240 mg / kg / day which can be increased to 300 mg / kg / day. The daily dose is administered at least in 3 IV injections spaced 8 hours apart.
. Preterm neonates and term neonates : the average dose is 160 mg / kg / day, which can be increased to 200 mg / kg / day. The daily dose is administered in 2 IV injections spaced 12 hours apart.
- In patients with renal insufficiency:
. Dosage adjustment should only be performed in the presence of renal impairment with creatinine clearance less than 40 ml / min. It is recommended to use a dosage equal to one third of the normal dosage.
. In severe renal impairment (creatinine clearance less than 10 ml / min), a loading dose of 80 mg / kg and then 40 mg / kg every 12 hours.
- In hemodialysis patients:
. The dialysance of mezlocillin is 35-40%. Also, a loading dose of 80 mg / kg then 40 mg / kg should be given every 12 hours until the next session.
. In peritoneal dialysis, same dosing regimen.
. In addition, it is possible to add 200 mg / L of mezlocillin to the dialysis liquid.
This medicine MUST NEVER BE USED in case of allergy to antibiotics of the beta-lactam family (penicillins, cephalosporins): take into account the risk of cross allergy with antibiotics of the cephalosporin group.
NOT RECOMMENDED :
This medicine SHOULD NOT BE USED in combination with methotrexate (see interactions).

- Infections and infestations:
- Gastrointestinal disorders:
. Nausea, vomiting, diarrhea.
. Pseudomembranous colitis (see warnings and precautions for use).
. Taste disorders.
- Immune system disorders:
Rash (exanthema, urticaria), pruritus, anaphylactic reactions including anaphylactic shock (see warnings and precautions for use).
- Skin and subcutaneous tissue disorders:
. Maculopapular rash of allergic origin or not.
. Stevens-Johnson syndrome, Lyell syndrome.
- General disorders and anomalies at the site of administration:
Pain or irritation at the injection site and more rarely thrombophlebitis, drug fever.
- Hematological and lymphatic system disorders:
Eosinophilia, anemia, leukopenia, agranulocytosis, reversible thrombocytopenia.
- Renal and urinary disorders:
Acute interstitial nephritis.
- disorders of the nervous system:
Convulsions (see warnings and precautions for use).
- Metabolism and nutrition disorders:
Transient elevation of transaminases (SGOT, SGPT), alkaline phosphatase, bilirubin, creatinine.
